Experimental research on analgesic effect and side-effects of centipede-scorpion powder in mice

摘要: 目的 研究蜈蚣全蝎散(CS)对小鼠的镇痛作用及伴随的毒副作用,为其"功效-毒性"相关性研究提供实验依据。 方法 将雄性昆明小鼠随机分为4组:生理盐水对照组(NS组)、CS 20 mg/kg组(C1组)、50 mg/kg组(C2组)、80 mg/kg组(C3组),不同剂量的CS灌胃,1次/d,连续4 d后,采用醋酸扭体法检测其镇痛效应;采用转棒试验检测小鼠感觉、运动行为改变;采用自主活动能力测试检测小鼠中枢神经系统功能状态改变;检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性改变,评价CS对小鼠肝毒性的影响。 结果 与对照组小鼠扭体数[(20.3±5.3)次/10 min]比较,各CS给药组小鼠扭体次数减少[分别为(11.8±6.5、7.6±5.7、4.1±2.8)次/10 min],抑制率分别为41.9%、62.6%、79.8%,其镇痛效果呈现剂量依赖性(P<0.05);在小鼠感觉、运动功能及自主活动能力测试中,与对照组比较,各浓度CS对小鼠感觉、运动功能、自主活动次数均无显著影响,差异均无统计学意义(P﹥0.05);肝毒性实验检测显示,与对照组血清ALT[(30.3±5.3) U/L]、AST[(19.5±3.7) U/L]比较,C3组小鼠血清ALT[(43.9±6.8) U/L]、AST[(32.8±5.8) U/L]活性升高,差异均有统计学意义(P<0.05)。 结论 蜈蚣全蝎散对实验小鼠具有剂量依赖性镇痛作用,并且低、中剂量对神经系统无明显影响,但高剂量能引起肝毒性损伤。Abstract: Objective To study the analgesic effects and side effects of Centipede-scorpion (CS) powder in mice so as to provide the experimental basis for efficacy-toxicity correlation. Methods Male Kunming mice were randomly divided into 4 groups:saline control group (group NS) and CS 20 mg/kg (group C1), CS 50 mg/kg (group C2) and CS 80 mg/kg (group C3), receive different doses of CS or NS for 4 days by gavage. The acetic acid writhing tests were adopted to observe analgesic effect by recording the mice writhing times. The rotarod performance test was used to examine the sensory and motor coordination of mice. Autonomic activity test was used for detecting the functional status of the central nervous system in mice. The levels of ALT and AST in serum were tested to evaluate its hepatotoxicity. Results Compared with the control group (20.3±5.3), the times of writhing was decreased in CS group (11.8±6.5, 7.6±5.7, 4.1±2.8, respectively), inhibition rates of analgesic were 41.9%, 62.6%, 79.8%, respectively, and the analgesic effect was dose-dependent (P<0.05). Compared with the control group, the average of the behavior of sensory and motor times in the rotarod performance test and autonomic activity test, were decreased in high-dose groups (P<0.05), the average of autonomic activity times were increased significantly in high-dose groups (P<0.05). Compared with the levels of ALT (30.3±5.3) and AST (19.5±3.7) in the control group, hepatotoxicity test showed that the activities of ALT (43.9±6.8) and AST (32.8±5.8) in serum were increased in C3 group (P<0.05). Conclusion Centipede-scorpion powder had analgesic effect on experimental mice, and at the same time, accompanied by neurological damage and hepatotoxicity in high- dose group.
