Effects of taurine on endoplasmic reticulum stress in rats with acute stress skeletal muscle injury

摘要: 目的 研究牛磺酸对离心运动大鼠骨骼肌中SOD、MAD、GRP78、GADD153的影响,以期解释牛磺酸通过减少内质网氧化应激保护骨骼肌损伤的机制。 方法 选取成年SD大鼠36只,分为空白对照1组(A组)、空白对照2组(B组)、1 d离心运动组(C组)、2 d离心运动组(D组)、1 d离心运动+牛磺酸组(E组)、2 d离心运动+牛磺酸组(F组)。培养后分别收集6组大鼠骨骼肌,采用黄嘌呤氧化酶法和硫代巴比妥酸法测定SOD、MAD含量,采用Western 印迹方法测定GRP78、GADD153表达,比较6组大鼠SOD、MAD、GRP78、GADD153的差异。 结果 A组、B组大鼠SOD含量高于C组、D组、E组、F组大鼠,与C组、D组大鼠比较差异具有统计学意义(均P<0.05),但与E组、F组大鼠比较差异无统计学意义(均P>0.05);E组、F组大鼠SOD含量高于C组、D组大鼠,差异无统计学意义(均P>0.05)。C组、D组大鼠MDA含量高于A组、B组、E组、F组大鼠(均P<0.05)。A组、B组、E组、F组大鼠SOD/MDA值均高于C组、D组大鼠(均P<0.05),E组、F组SOD/MDA值低于A组、B组(均P<0.05)。C组、D组大鼠GRP78、GADD153表达量高于A组、B组(均P<0.05),但低于E组、F组大鼠(均P<0.05);E组、F组大鼠GRP78、GADD153表达量高于A组、B组(均P<0.05)。 结论 牛磺酸可能是通过提高内质网的抗氧化应激能力,从而维持骨骼肌抗氧化能力,加快骨骼肌中氧自由基的清除,减少剧烈运动后机体骨骼肌的氧化应激损伤,有利于骨骼肌功能的恢复。Abstract: Objective To study the effect of taurine on GRP78, MAD, GADD153 and SOD in rats with eccentric exercise, in order to explain the mechanism of taurine protecting skeletal muscle injury by reducing endoplasmic reticulum oxidative stress. Methods A total of 36 adult SD rats were randomly divided into blank control 1 group(group A), blank control 2 group(group B), 1 day eccentric exercise group(group C), 2 days eccentric exercise group(group D), 1 day eccentric exercise+taurine group(group E), 2 days eccentric exercise+taurine group(group F). The skeletal muscle of 6 groups rats were collected, and the contents of SOD, MAD, GRP78 and GADD153 were measured. The content of SOD and MAD were measured by xanthine oxidase method and thiobarbituric acid method, respectively. The expressions of GRP78 and GADD153 were measured by Western blotting method. The differences of SOD, MAD, GRP78 and GADD153 among the 6 groups were compared. Results The SOD in group A and group B was higher than that in group C, group D, group E and group F, there was significant statistical difference compared with group C, group D(P<0.05), but there was no difference compared with group E and group F(P>0.05). The SOD in group E and group F was higher than that in group C and group D, there was no statistically significant difference(P>0.05). The MDA in group C and group D was higher than that in group A, group B, group E and group F(P<0.05). The MDA/SOD value in group A, group B, group E and group F was greater than group C and group D(P<0.05). The MDA/SOD value in group E and group F was lower than that in group A and group B(P<0.05). GRP78, GADD153 expression in group C and group D were higher than that in group A and group B(P<0.05), but was lower than that in group E and group F(P<0.05). GRP78, GADD153 expression in group E and group F were higher than that in group A and group B(P<0.05). Conclusion Taurine may be maintain the antioxidant capacity of skeletal muscle by improving the ability of endoplasmic reticulum oxidative stress, which can accelerate the elimination of oxygen free radicals in skeletal muscle and decrease oxidative stress in skeletal muscle after strenuous exercise. It is conducive to the recovery of skeletal muscle function.
