抑郁症患者外周血内质网应激相关基因及microRNA-16、microRNA-124和microRNA-195表达研究

摘要: 目的 探讨抑郁症患者外周血内质网应激相关基因及microRNA-16、microRNA-124和microRNA-195表达水平。 方法 选择杭州市第七人民医院2017年7月—2018年7月期间收治的抑郁症患者97例作为研究对象;另选择2017年7月—2018年7月期间健康体检者63例作为对照组。采用Western blotting检测IRE1α和JNK表达,根据OD相对值=目的条带表达强度/GAPDH表达强度,进行相对值计算统计分析;采用PT-cPCR检测microRNA-16、microRNA-124和microRNA-195表达,获得原始Ct值,取3次结果平均值,应用2-ΔCT方法计算miRNA相对值。 结果 抑郁组IRE1α和JNK表达OD相对值高于对照组,差异具有统计学意义(P<0.05)。抑郁组microRNA-16和microRNA-195表达水平低于对照组,而microRNA-124表达水平高于对照组,差异具有统计学意义(P<0.05)。 结论 抑郁症患者外周血内质网应激相关基因IRE1α和JNK呈高表达,且microRNA-16和microRNA-195表达水平异常降低,而microRNA-124表达水平异常升高,故而认为可作为诊断和评估患者预后的生物学指标。

Abstract: Objective To investigate the expression of endoplasmic reticulum stress-related genes and microRNA-16, microRNA-124 and microRNA-195 in peripheral blood of depressive patients. Methods A total of 97 patients with depression admitted to our hospital from July 2017 to July 2018 were selected as the study subjects, and 63 health examinees from July 2017 to July 2018 were selected as the control group. Western blot was used to detect the expression of IRE1 alpha and JNK, and the relative value was calculated and analyzed according to OD relative value = target band expression intensity/GAPDH expression intensity. PT-cPCR was used to detect the expression of microRNA-16, microRNA-124 and microRNA-195, and the original CT value was obtained. The average value of three times was taken and the relative value of microRNA was calculated by 2-CT method. Results The relative value of OD expression of IRE1α and JNK in depression group was higher than that in control group (P<0.05). The expression levels of microRNA-16 and microRNA-195 in depression group were lower than those in control group, while the expression levels of microRNA-124 in depression group were higher than those in control group (P<0.05). Conclusion IRE1 α and JNK are highly expressed in the peripheral blood of depressive patients, and the expression levels of microRNA-16 and microRNA-195 are abnormally decreased, while the expression levels of microRNA-124 are abnormally increased. Therefore, it is considered that IRE1 alpha and JNK can be used as biological indicators for diagnosis and evaluation of prognosis of depressive patients.-
