Activation of PKC δ related signal pathway factor and its effect on cell proliferation in human growth hormone type pituitary adenoma cells

摘要: 目的 分析蛋白激酶C(protein kinase C,PKCs)相关信号通路因子在人生长激素(growth hormone,GH)型垂体腺瘤细胞的表达及调节细胞增殖的作用机制。 方法 收集2016年5月—2018年3月蚌埠医学院第一附属医院收治的8例GH型垂体腺瘤患者术中切除肿瘤组织进行原代培养,免疫组化法检测PKCδ、CREB、ERK1/2在细胞内的表达。将原代培养的细胞分为对照组(空白)、激动剂组(PMA)、抑制剂组(Rottlerin)、联合组(PMA+Rottlerin),各8株细胞,ELISA法检测各组GH分泌水平;CCK-8法检测细胞活性;Western blotting检测各组因子及磷酸化水平,使用SPSS 21.0统计学软件进行数据分析。 结果 PKCδ、CREB、ERK1/2均较多见于GH型垂体腺瘤细胞质。与对照组比较,激动剂组GH分泌及细胞活性升高,抑制剂组明显降低(均P<0.01),联合组GH分泌及细胞活性低于激动剂组,高于抑制剂组(均P<0.01)。与对照组比较,激动剂组p-PKCδ、p-CREB、p-ERK1/2水平升高,抑制剂组三者水平降低(均P<0.01),联合组p-CREB、p-ERK1/2水平低于激动剂组,高于抑制剂组(均P<0.01)。 结论 PKCδ、CREB、ERK1/2在GH型垂体腺瘤细胞中存在明显高表达,激活PKCδ表达可提高p-CREB水平促进GH分泌,同时提高p-ERK1/2水平促进细胞增殖。

Abstract: Objective To analyze the expression of protein kinase C(PKCs) related signal pathway factors in human growth hormone(GH) type pituitary adenoma cells and its mechanism of regulating cell proliferation. Methods Eight patients with GH type pituitary adenoma were treated with resection of tumor tissue for primary culture. The expression of PKCδ, CREB and ERK1/2 in the cells was detected by immunohistochemistry. The primary cultured cells were divided into control group(blank), agonist group(PMA), inhibitor group(rottlerin), combination group(PMA+Rottlerin), 8 cells in each group. The GH secretion level of each group was detected by ELISA, the cell activity was detected by CCK-8, the factors and phosphorylation level were detected by western blot. The data was analyzed by SPSS 21.0 software package. Results PKCδ, CREB and ERK1/2 were more common in the cytoplasm of GH pituitary adenoma. Compared with the control group, GH secretion and activity increased in the agonist group, but significantly decreased in the inhibitor group(all P<0.01). Compared with the control group, the levels of p-PKCδ, p-CREB and p-ERK1/2 in the agonist group increased, while those in the inhibitor group decreased(all P<0.01). The levels of p-CREB and p-ERK1/2 in the combined group were lower than those in the agonist group and higher than those in the inhibitor group(all P<0.01). Conclusion PKCδ, CREB and ERK1/2 are highly expressed in GH pituitary adenoma cells. Activation of PKCδ can increase the level of p-CREB and GH secretion, and increase the level of p-ERK1/2 to promote cell proliferation.-
