Study on expressions of GAS5 and PTEN in serum of elderly patients with hypertension and their relationships with endothelial function

摘要: 目的 通过检测老年高血压患者血清中长链非编码RNA(long non-coding RNA,lncRNA)生长特异抑制物5(growth arrest-specific 5,GAS5)和人类第10号染色体缺失磷酸酶及张力蛋白同源基因(phosphatase and tensin homologues deleted on human chromosome ten,PTEN)蛋白表达水平,分析二者与内皮功能的关系。 方法 选择2018年5月—2019年5月金华市中心医院收治的老年高血压患者87例,另选择同期体检健康者87例作为对照组。实时荧光定量(qRT-PCR)法检测lncRNA GAS5表达水平,ELISA法检测PTEN蛋白表达水平。使用电子血压计测定收缩压(SBP)和舒张压(DBP),使用超声诊断仪检测内皮依赖性血管扩张率(FMD),分析GAS5、PTEN水平与以上指标相关性。 结果 老年高血压患者收缩压、舒张压显著高于对照组(均P<0.05)。老年高血压患者vWF水平显著高于对照组(P<0.05),老年高血压组患者FMD值、NO水平显著低于对照组(均P<0.05)。老年高血压患者血清lncRNA GAS5及PTEN蛋白水平显著低于对照组(均P<0.05)。老年高血压患者血清lncRNA GAS5及PTEN蛋白水平与NO水平呈正相关(r=0.563,P=0.005;r=0.507,P=0.006),与vWF呈负相关(r=-0.457,P=0.004;r=-0.426,P=0.008)。老年高血压患者血清GAS5与PTEN水平呈正相关(r=0.484,P<0.001)。 结论 GAS5与PTEN在老年高血压患者血清中表达下调,与患者血管内皮细胞损伤、血管舒张功能降低有关。

Abstract: Objective To analyze the relationships between lncRNA GAS5 and PTEN protein expressions and endothelial function in elderly patients with hypertension. Methods A total of 87 elderly hypertensive patients admitted to Jinhua Central Hospital from May 2018 to May 2019 were selected, and another 87 healthy persons were selected as control group during the same period. Real-time fluorescence quantitative(qRT-PCR) was used to detect the expression of lncRNA GAS5, and ELISA was used to detect the expression of PTEN. Meanwhile, the change rate of vascular diameter(FMD), systolic blood pressure(SBP) and diastolic blood pressure(DBP) were measured by electronic sphygmomanometer, and endothelium-dependent vasodilation rate(FMD) was measured by ultrasound diagnostic instrument, and the correlations between GAS5 and PTEN levels and the above indexes were analyzed. Results Systolic blood pressure and diastolic blood pressure in elderly patients with hypertension were significantly higher than those in the control group(all P<0.05). The level of vWF in elderly patients with hypertension was significantly higher than that in control group(P<0.05), and the FMD value and NO level in elderly patients with hypertension were significantly lower than those in control group(all P<0.05). The levels of serum lncRNA GAS5 and PTEN protein in elderly patients with hypertension were significantly lower than those in the control group(all P<0.05). Serum lncRNA GAS5 and PTEN protein levels were positively correlated with NO levels in elderly patients with hypertension(r=0.563, P=0.005; r=0.507, P=0.006), and negatively correlated with vWF(r=-0.457, P=0.004; r=-0.426, P=0.008). There was a positive correlation between serum GAS5 and PTEN level in elderly patients with hypertension(r=0.484, P<0.05). Conclusion The expressions of GAS5 and PTEN in serum of elderly patients with hypertension are down-regulated, which are related to the injury of vascular endothelial cells and the decrease of vascular relaxation function.
