Correlation between lncRNA HOTAIR and tumor markers and pathological stage of lung cancer

摘要: 目的 观察肺癌不同病理分期lncRNA HOTAIR及肿瘤标志物变化水平,探讨lncRNA HOTAIR联合肿瘤标志物诊断肺癌分期的临床价值。 方法 将2019年1—9月蚌埠医学院第一附属医院收治的50例肺癌患者作为肺癌组,50例健康志愿者为对照组,根据肺癌T、N分期可分为T1、T2、T3期组和N0、N1、N2期组。荧光原位杂交检测癌组织和癌旁组织lncRNA HOTAIR水平;免疫荧光法检测受试者血清NSE、CEA、CYFRA21-1的水平。 结果 癌旁组织中lncRNA HOTAIR浓度[(0.98±0.04)ng/mL]低于肺癌组织[(3.56±0.15)ng/mL,t=13.706,P<0.001];肺癌组血清NSE、CEA、CYFRA21-1水平均高于对照组(均P<0.05);肺癌T、N分期分组中,临床期数越高,lncRNA HOTAIR、NSE、CEA和CYFRA21-1表达水平均随之升高(均P<0.05);lncRNA HOTAIR表达与NSE、CEA、CYFRA21-1表达呈正相关(均P<0.05);肺癌患者组织lncRNA HOTAIR和血清NSE、CEA、CYFRA21-1表达水平与肺癌T分期均呈正相关(均P<0.05),与肺癌N分期均呈正相关(均P<0.001);联合检测的灵敏度、约等指数和阳性预测值均高于单一指标。 结论 lncRNA HOTAIR和血清NSE、CEA、CYFRA21-1表达水平与肺癌病理分期密切相关,lncRNA HOTAIR联合血清肿瘤标志物判断肺癌病理分期可提高结果的准确性。

Abstract: Objective To observe the changes of lncRNA HOTAIR and tumor markers in different pathological stages of lung cancer, and to explore the clinical value of lncRNA HOTAIR combined with tumor markers in the diagnosis of lung cancer stages. Methods Fifty patients with lung cancer in the first affiliated Hospital of Bengbu Medical College were selected as the lung cancer group and 50 healthy volunteers as the control group. According to the T and N stages of lung cancer, 50 patients with lung cancer were divided into T1, T2, T3 and N0, N1, N2 groups. The levels of lncRNA HOTAIR in cancer and paracancerous tissues were detected by fluorescence in situ hybridization, and the levels of NSE, CEA and CYFRA21-1 in serum were detected by immunofluorescence. Results The serum levels of NSE, CEA and CYFRA21-1 in the lung cancer group were higher than those in the control group, and the expression levels of, lncRNA HOTAIR, NSE, CEA and CYFRA21-1 in the lung cancer group were significantly higher than those in the control group(all P<0.05). In the T and N stages of lung cancer, the expression levels of, lncRNA HOTAIR, NSE, CEA and CYFRA21-1 were significantly higher than those in the control group(all P<0.05). LncRNA HOTAIR concentration in paracancerous tissues was lower than that in lung cancer tissues(P<0.001), and the serum levels of NSE, CEA and CYFRA21-1 in lung cancer group were higher than those in control group(all P<0.05). The expression of lncRNA HOTAIR was positively correlated with the expression of NSE, CEA and CYFRA21-1. The expression levels of lncRNA HOTAIR in tissue and serum NSE, CEA and CYFRA21-1 in patients with lung cancer were positively correlated with T stage and N stage of lung cancer(all P<0.001), and were positively correlated with the N stage of lung cancer, and the expression levels of tissue and serum NSE, CEA, CYFRA21-1 in patients with lung cancer were positively correlated with T stage and N stage of lung cancer(all P<0.001). The sensitivity, approximate equal index and positive predictive value of combined detection were higher than those of single index. Conclusion The expression level of lncRNA HOTAIR and serum NSE, CEA, CYFRA21-1 is closely related to the pathological stage of lung cancer. lncRNA HOTAIR combined with serum tumor markers can improve the accuracy of detection of pathological stage of lung cancer.-
