Expression and significance of placenta-specific protein 1 mRNA in placenta and peripheral blood of patients with fetal growth restriction

摘要: 目的 本文通过测定及分析胎盘特异性蛋白1(placenta-specific protein 1,PLAC1)在胎儿生长受限(fetal growth restriction,FGR)孕妇胎盘和外周血中的异常表达情况,对其意义进行探讨。 方法 选择嘉兴市妇幼保健院2017年4月—2020年1月住院分娩的FGR孕妇40例作为FGR组,同期住院分娩的40例正常孕妇作为对照组。测量胎盘厚度、胎盘质量、新生儿体重。采用逆转录-聚合酶链反应(RT-PCR)测定胎盘和外周血中PLAC1 mRNA水平。采用t检验分析2组之间各指标的差异,采用Pearson分析指标之间的相关性。 结果 FGR组胎盘厚度、胎盘质量、新生儿体重均低于对照组(均P<0.05)。FGR组胎盘中PLAC1 mRNA水平(0.47±0.09)低于对照组(1.00±0.07,t=29.399,P<0.05),外周血中PLAC1 mRNA水平(1.54±0.11)高于对照组(1.00±0.08,t=25.109,P<0.05)。FGR孕妇胎盘PLAC1 mRNA水平与胎盘厚度、胎盘质量、新生儿体重均呈正相关(均P<0.05)。FGR孕妇外周血中PLAC1 mRNA水平与胎盘厚度、胎盘质量、新生儿体重均呈负相关(均P<0.05)。FGR孕妇胎盘PLAC1 mRNA水平与外周血中PLAC1 mRNA水平呈负相关(r=-0.602,P<0.001)。 结论 检测胎盘及外周血中PLAC1 mRNA的水平,可一定程度上反映FGR的严重程度。Abstract: Objective To detect the abnormal expression of placenta-specific protein 1(PLAC1) of patient with fetal growth restriction(FGR), and discuss its significance. Methods Forty FGR pregnant women who were hospitalized and delivered in Jiaxing Maternal and Child Health Hospital from April 2017 to January 2020 were selected as the FGR group, and 40 normal pregnant women who were hospitalized and delivered during the same period were selected as the control group. The placental thickness, placental mass and neonatal weight were measured. Reverse transcription-polymerase chain reaction(RT-PCR) was used to determine PLAC1 mRNA levels in placenta and peripheral blood. The t-test was used to analyze the differences of the indicators between the two groups. Correlation between indicators was analyzed using Pearson. Results The placental thickness, placental mass and neonatal weight in the FGR group were lower than those in the control group(all P<0.05).The level of PLAC1 mRNA in the placenta of the FGR group(0.47±0.09) were lower than that of the control group(1.00±0.07), t=29.399, P<0.05.The level of PLAC1 mRNA in the peripheral blood of the FGR group(1.54±0.11) was higher than that of the control group(1.00±0.08), t=25.109, P<0.05. The placental PLAC1 mRNA level in FGR pregnant women was positively correlated with placental thickness, placental mass and neonatal weight(all P<0.05). The peripheral blood PLAC1 mRNA level in FGR pregnant women were negatively correlated with placental thickness, placental mass and neonatal weight(all P<0.05).There was negatively correlation between PLAC1 mRNA levels in the placenta and peripheral blood of FGR pregnant women(r=0.602, P<0.001). Conclusion The level of PLAC1 mRNA in placenta and peripheral blood can reflect the severity of FGR.
