Expression of pDC,TLR7 and TLR9 in labial salivary glands of patients with in primary Sjögren’s syndrome

摘要: 目的 检测浆细胞样树突状细胞(plasmacytoid dendritic cell,pDC),Toll样受体7(Toll-like receptor 7,TLR 7),Toll样受体9(Toll-like receptor 9,TLR 9)在原发性干燥综合征患者唇腺组织中的表达情况。 方法 采用免疫组织化学法检测2018年1月—2019年12月昆明医科大学第二附属医院风湿免疫科门诊及住院部25例确诊为原发性干燥综合征的女性患者唇腺组织中pDC特异性抗原CD123及TLR7、TLR9的表达,在低倍镜下(10×20)随机选取4个视野利用Image-Pro Plus 6.0软件和SPSS 17.0软件进行统计分析。 结果 pSS患者唇腺组织中可见pDC、TLR7、TLR9阳性细胞主要分布于主要分布于腺泡、腺导管上皮细胞、淋巴细胞浸润灶。CD123+ pDC表达(3.47±0.26)%高于正常对照组(2.36±0.15)%;TLR7(2.06±0.37)%高于正常对照组(0.14±0.05)%;TLR9(1.80±0.26)%高于正常对照组(0.84±0.08)%,差异均有统计学意义(均P<0.05)。 结论 pDC、TLR 7、TLR 9在pSS患者的唇腺组织(腺导管和腺泡的上皮细胞)及淋巴细胞浸润灶内高表达。Abstract: Objective To detect the expression of plasma cell dendritic cell(pDCs), Toll-like receptor 7(TLR7), Toll-like receptor 9(TLR9) in labial glands of patients with primary Sj9 gren's syndrome(pSS). Methods The expression of pDC specific antigen CD123, TLR7 and TLR9 in labial gland tissues of 25 female patients diagnosed as pSS in the outpatient and inpatient departments of the Department of Rheumatology and Immunology of the Second Affiliated Hospital of Kunming Medical University from January 2018 to December 2019 were detected by immunohistochemical method. Four visual fields were randomly selected under low magnification(10×20) and analyzed by Image pro plus 6.0 software and SPSS 17.0 software. Results pDC, TLR7 and TLR9 positive cells were mainly distributed in acinar, ductal epithelial cells and lymphocyte infiltration foci in labial glands of patients with pSS. The expression of CD123+ pDC(3.47±0.26)% was higher than that of the normal control group(2.36±0.15)%, TLR7(2.06±0.37)% was higher than that of the normal control group(0.14±0.05)%, TLR9(1.80±0.26)% was significantly higher than that of the normal control group(0.84±0.08)%, all P<0.05. Conclusion pDC, TLR7 and TLR9 are highly expressed in labial glands(epithelial cells of glandular ducts and acini) and infiltrating foci of lymphocytes in patients with pSS.
