Fibulin-1 expression in tongue cancer and clinical value based on bioinformatics analysis

  • 摘要:   目的  探讨舌癌组织及舌癌细胞中纤维蛋白1(FBLN1)的表达及临床意义。  方法  基于HPA数据库分析FBLN1在头颈部癌组织中的表达情况;GEPIA数据库和Pearson相关性分析研究FBLN1与MKI67两者在头颈部癌中表达的关联;Kaplan-Meier Plotter数据库在线分析FBLN1表达水平与头颈部癌患者生存率的关系;免疫组化和Western blotting实验验证FBLN1在舌癌组织中的表达情况;构建过表达FBLN1舌癌细胞株,通过划痕实验、Transwell实验检测FBLN1对舌癌细胞迁移、侵袭能力的影响。  结果  HPA数据库检索和免疫荧光显示,FBLN1在人体正常细胞和组织中高表达,在头颈部癌组织中较癌旁组织低表达(P<0.05)。GEPIA数据库分析显示,FBLN1在头颈部癌与癌旁组织中表达水平差异无统计学意义(P>0.05),而MKI67在头颈部癌中高表达(P<0.05),且Pearson相关性分析显示二者表达不相关(P=0.280)。Kaplan-Meier Plotter生存分析表明,FBLN1表达与头颈部癌总生存率无关(log-rank P=0.190),而FBLN1高表达时无病生存率更高(log-rank P=0.007)。免疫组化结果显示,FBLN1在癌旁组织中的染色强度和阳性面积均高于肿瘤组织(P<0.001)。Western blotting结果显示,在10例舌癌及配对癌旁组织中,FBLN1蛋白水平在癌旁组织中表达更高(P<0.001)。划痕实验和Transwell实验结果显示,过表达FBLN1显著抑制舌鳞癌细胞Cal-27的迁移、侵袭能力(P<0.05)。  结论  FBLN1基因在舌癌组织中低表达,而过表达FBLN1抑制舌鳞癌细胞Cal-27的迁移和侵袭能力,提示其可作为评价舌癌患者预后的潜在标志物。
