湿疹与抑郁症的孟德尔随机化研究

Mendelian randomization study of eczema and depression

  • 摘要:   目的  湿疹与抑郁症之间的关系备受关注,然而二者之间的因果关联尚不明确。本研究采用孟德尔随机法(MR)研究湿疹与抑郁症之间是否存在因果关系。  方法  在公开的全基因组关联研究(GWAS)的集合数据集中选出湿疹与抑郁症的基因型数据,湿疹为暴露因素,抑郁症为结局变量。样本量为40 835例。利用MR对所选样本进行基因型分析,其中工具变量为与湿疹疾病有强相关性的基因位点。采用比值比(OR)作为估计指标,效应值β判断两者之间的关联方向。同时进行异质性检验、敏感性分析以及水平多效性检验。  结果  逆方差加权法分析结果显示,湿疹与抑郁发生之间呈正相关关系(OR=1.011,95% CI:1.001~1.022,P=0.037);加权中位数回归分析得到了相似结果(OR=1.012,95% CI: 1.001~1.023,P=0.029)。Cochran's Q检验显示结果存在显著异质性(P < 0.05),但通过随机效应模型分析得到P=0.037且β=0.011,证明二者存在正向因果关系。此外,漏斗图和Mr-Egger截距未观察到水平多效性。  结论  研究结果表明,湿疹与抑郁发生之间可能存在正向因果关系。
