An experimental study of effect of Wenshen Yiqi medicine on expression of MMP-2 and MMP-9 in rats with severe pneumonia

摘要: 目的 观察温肾益气中药对重症肺炎大鼠金属基质蛋白酶(MMP-2、MMP-9)表达的影响。 方法 将100只SPF级SD大鼠随机分为5组:对照组正常饲养,其余4组气管穿刺注入肺炎克雷伯菌制备重症肺炎模型,并分别以无菌蒸馏水及相应浓度的温肾益气饮灌胃,1次/d,共7 d。观察肺组织病理变化,并采用血常规分析仪测定支气管肺泡灌洗液白细胞和中性粒细胞计数,采用免疫蛋白印迹实验测定肺组织PI3K、AKT1、P-AKT1、MMP-2、MMP-9的蛋白表达水平。 结果 与对照组相比,其余各组明显偏高(P<0.05),与模型组相比,中剂量组及高剂量组均有所降低(P<0.05),低剂量组无明显变化。各组大鼠之间的PI3K和AKT1蛋白表达水平差异无统计学意义,但P-AKT1、MMP-2及MMP-9明显不同(P<0.05);与对照组相比,其余各组明显升高(P<0.05),与模型组相比,低剂量组、中剂量组及高剂量组均有明显降低,且各用药组之间的差异也均有统计学意义(P<0.05)。MMP-2和MMP-9与P-AKT1表达水平呈正相关性(r=0.896、0.547,P<0.05)。 结论 温肾益气中药能够通过PI3K/AKT通路下调MMP-2与MMP-9的表达,减轻细胞外基质降解和炎性应激反应,可能为其治疗重症肺炎的药理机制之一。

Abstract: Objective To observe the effect of Wenshen Yiqi medicine on the expression of matrix metalloproteinases(MMP-2,MMP-9) in rats with severe pneumonia. Methods Total 100 SPF SD rats were randomly divided into five groups:rats in control group were given normal feeding,and rats in the other four groups made severe pneumonia model by tracheal puncture injection model were given sterile distilled water and corresponding concentration of Wenshen Yiqi drink,1 times/d,treatment for 7 d.The Lung histopathological changes were observed,leukocyte and neutrophils of bronchoalveolar lavage fluid were accounted by Blood Analyzer,and the expression of lung tissue PI3K,AKT1,P-AKT1,MMP-2 and MMP-9 were detected by Western blot test. Results When compared with control group,the markers of other groups were significantly higher(P<0.05);compared with model group,the markers of middle and high dose group were significantly lower(P<0.05),but there was no significantly different with low dose group.There were no significantly different in PI3K and AKT1 protein expression between five groups(P>0.05).But there were significantly different in P-AKT1,MMP-2 and MMP-9 expression between five groups(P<0.05);As compared with control group,the markers of other groups were significantly higher(P<0.05);as compared with model group,the markers of low,middle and high dose group were significantly lower (P<0.05),and there were significantly different in each dose group(P<0.05).The relationship of MMP-2 and MMP-9 with P-AKT1 were positive correlation. Conclusion Wenshen Yiqi method can effectively reduce the expression of MMP-2 and MMP-9 through PI3K/AKT signaling pathway,reduce the degradation of extracellular matrix and inflammatory response,which might be one of the pharmacological mechanism for the treatment of severe pneumonia.
