Effects of coix seed triglyceride injection combined with gemcitabine on the proliferation and apoptosis of Bel-7402

摘要: 目的 研究薏苡仁注射液(coix seed triglyceride)联合吉西他滨(gemcitabine)对人肝癌细胞株Bel-7402增殖及凋亡的影响及可能机制。 方法 本实验以体外培养的人肝癌细胞株Bel-7402为研究对象,采用四甲基偶氮唑蓝(MTT)法、克隆形成实验测定薏苡仁、吉西他滨及两药联合作用对人肝癌细胞株Bel-7402活性及增殖能力的影响,流式细胞仪测定凋亡率,逆转录聚合酶链反应(RT-PCR)测定Caspase-3、Bcl-2表达水平,Image-pro plus 6.0计数克隆形成细胞集落数。使用SPSS 17.0统计学软件对实验结果进行分析。 结果 薏苡仁及吉西他滨对人肝癌细胞株Bel-7402活性及增殖均有抑制作用,两者联合具有协同作用(F=509.677,P<0.01;F=562.100,P<0.01)。薏苡仁联合吉西他滨对人肝癌细胞株Bel-7402的凋亡率为(76.3±1.6)%,明显高于吉西他滨的单药作用[(53.2±2.0)%]及薏苡仁[(42.4±1.8)%]的单药作用(F=46.742,P<0.01)。两药联合组细胞Caspase-3表达增加(F=38.264,P<0.01),Bcl-2表达量降低(F=4.817,P<0.05)。 结论 薏苡仁可能通过影响细胞凋亡相关因子如Caspase-3、Bcl-2抑制肝癌细胞活性及增殖,诱导细胞凋亡,起到对吉西他滨化疗增敏的作用。Abstract: Objective To investigate effects and mechanism of coix seed triglyceride combined with gemcitabine on proliferation and apoptosis of Bel-7402. Methods The MTT and colony formation assay were used to detect cell inhibition and cell proliferation.Flow cytometry was used to detect the apoptosis of cells.The expression of Caspase-3 and Bcl-2 were detected by reverse transcriptase polymerase chain reaction(RT-PCR) in different group.Image-Pro Plus 6 was used to count the number of colony forming clones.SPSS 17.0 was used to analyze the experimental results. Results The cell viability and proliferation was inhibited by coix seed triglyceride and gemcitabine.And the synergistic effect of cell viability and proliferation was observed when coix seed triglyceride and gemcitabine were administered together(F=509.677,P<0.01;F=562.100,P<0.01).The apoptosis rate of Bel-7402 cells treated with coix seed triglyceride combined with gemcitabine was significantly higher than those treated with coix seed triglyceride or gemcitabine alone[(76.3±1.6)%,(53.2±2.0)%,(42.4±1.8)%,F=46.742,P<0.01].The expression of Caspase-3 was increased(F=38.264,P<0.01),but the expression of Bcl-2 was decreased(F=4.817,P<0.05) in combination group. Conclusion The inhibition of cell viability and proliferation of Bel-7402 cells can be synergistically enhanced and the rate cells apoptosis was increased by the treatment of coix seed triglyceride combined with gemcitabine,which provides reference for the clinical application of the two drugs.
