Effects of miR-130a on EPCs adhesion, migration, and expressions of IL-18 and TGF-β1 in peripheral blood of patients with type 2 diabetes mellitus

  • Objective To study the effects of miR-130 a on the adhesion and migration abilities of endothelial progenitor cells(EPCs) and the expressions of interleukin-18(IL-18) and transforming growth factor-β1(TGF-β1) in peripheral blood of patients with type 2 diabetes mellitus. Methods Peripheral blood of 60 patients with type 2 diabetes mellitus in Wenzhou Hospital of Traditional Chinese Medicine from April 2018 to August 2018 was collected. EPCs were isolated, cultured and identified by fluorescence staining. EPCs were divided into three groups:control group, mimics NC group, and miR-130 a mimics group. The control group were cultured without any treatment, the mimics NC group were transfected with mimics NC, and the miR-130 a mimics group were transfected with miR-130 a mics. The expression of miR-130 a was detected by qRT-PCR. The effects of overexpression of miR-130 a on the adhesion and migration of EPCs were observed. The expressions of IL-18 and TGF-β1 in the cell culture supernatant was detected by ELISA. Results After 48 h of culture, the cells grew adherently, and were round and small. On the 4 th day, the cell volume increased and was elliptical. On the 7 th day, the cells adhered to each other and formed clusters, the cells were round or irregular. The cells were positive for Dil-acLDL, FITC-UEA-I, and DAPI staining, which confirmed EPCs. There were no significant differences in the relative expression of miR-130 a, the numbers of adhesion and migration of EPCs, and the expression levels of IL-18 and TGF-β1 between the control group and the mimics NC group(all P>0.05). Compared with the control group and the mimics NC group, the relative expression of miR-130 a in EPCs of the mimics group increased significantly(all P<0.05), the numbers of adhesion and migration of EPCs increased(all P<0.05), the expression levels of IL-18 and TGF-β1 decreased(all P<0.05). Conclusion Overexpression of miR-130 a can improve the adhesion and migration of EPCs in peripheral blood of patients with type 2 diabetes mellitus, and inhibit the expressions of IL-18 and TGF-β1.
