  •   Objective  To identify the potential hub genes for the prognosis and targeted therapy of stomach cancer.  Methods  GSE33651 and GSE118916 datasets were selected from gene expression omnibus (GEO, study periods were from November 2011 to August 2019). The differentially expressed genes (DEGs) were identified by GEO2R. Gene ontologty (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses of DEGs was performed in DAVID database. Protein-protein interaction (PPI) network was constructed using STRING database and Cytoscape software. DEGs with Degree >10 were considered as hub genes. Based on gastric adenocarcinoma data from the cancer genome atlas (TCGA), survival and expression analyses of hub genes were performed in OncoLnc and UALCAN databases, respectively. Immune infiltration analysis of hub genes was performed using TIMER database.  Results  A total of 80 up-regulated and 34 down-regulated DEGs were identified, enriched in 69 GO terms and 7 KEGG pathways.Fourteen hub genes were screened from the PPI network. The expression levels of FN1, COL4A2 and COL4A1 in gastric cancer tissue were higher than those in normal gastric tissues, and showed excellent prognostic values in gastric cancer. FN1, COL4A2 and COL4A1 were closely associated with immune cell infiltration, which might be involved in the immune response of gastric cancer.  Conclusion  FN1, COL4A2 and COL4A1 are the hub genes of gastric cancer, which may be potential prognostic biomarkers and therapeutic targets for gastric cancer.
