Objective To analyze the impact of Xuezhikang on suppressor of cytokine signaling 3 (SOCS-3) mRNA expression levels in obese rats liver,and explore theregulatory mechanism of Xuezhikang on glucose and lipid metabolism.
Methods The obese rats model were established and divided into model group,low dose Xuezhikang group (Xuezhikang 0.23 g/kg ),middle dose Xuezhikang group (Xuezhikang 0.69 g/kg) and high dose Xuezhikang group (Xuezhikang 1.15 g/kg),with 10 rats in each group,all the rats in the four groups were administrated with high-fat foods;10 rats to which no Xuezhikang was applied were selected as null group and were administrated with conventional food.The rats in null group and model group received the same volume of water as Xuezhikang.The levels of serum insulin,leptin,glucose,C-peptide and liver SOCS-3 mRNA were detected.
Results The levels of serum insulin,leptin,glucose and C-peptide in the model group were significantly higher as compared with the null group(
P<0.05),but they were decreased by Xuezhikang treatment(positively with the dose of Xuezhikang),
P<0.05;The liver SOCS-3 expression was significantly increased as compared with the null group(
P<0.05),rat liver SOCS-3 mRNA expression was depressed by Xuezhikang treatment (positively with the dose of Xuezhikang),
P<0.05;The level of liver SOCS-3 mRNA was negative correlated with serum leptin(
r=-0.612) and serum insulin(
r=-0.726),
P<0.05.
Conclusion Xuezhikang can effectively decrease the levels of serum glucose,leptin,insulin and C-peptide and improve lipid disorders through regulating the expression of SOCS-3 mRNA.
