Objective To explore the expression of NPAS2 in AML patients and cell lines, and to analyze the effect of NPAS2 on the growth of AML cells.
Methods The mRNA expression of NPAS2 in 46 AML patients was analyzed from 2013 to 2017 by real-time PCR. The protein expression of NPAS2 in AML cell lines was analyzed by western blot. The relationship between the expression of NPAS2 and prognosis of AML patients was analyzed by statistical method. The effects of NPAS2 on the growth of AML cells were detected by using CCK-8 after using RNAi to interfere the expression of NPAS2.
Results The expression of NPAS2 in AML patients and cell lines was higher than that in control group (
P<0.05). Compared with control group, the expression level of NPAS2 in the AML-M1, M2, M3, M4 and M5 patients was higher (
P<0.05), and the expression level in the AML-M6 and M7 patients was no different (
P>0.05). AML patients with high expression of NPAS2 had a shorter overall survival period (
P<0.05). Compared with the control group, the cell growth of the RNAi-NPAS2 group was significantly decreased in AML cells (
P<0.05).
Conclusion The expression NPAS2 was raised in AML patients and negatively correlated with the prognosis of AML patients, while NPAS2 promoted AML cell growth.
