Objective To explore the effects of fasting blood glucose(FBG) ≥ 5.6 mmol/L plus CC genotypes of rs5417 locus in Glut4 and of FBG ≥ 5.6 mmol/L on the development of obstructive sleep apnea syndrome(OSAS).
Methods Eight hundred and seventy hypertensive inpatients,who were highly suspicious of OSAS and underwent nocturnal polysomnography from January to December 2010,were available.Two hundred and four subjects had a FBG ≥ 5.6 mmol/L,of whom,non-relative 96(84 males) subjects were selected to perform sequencing of functional regions of Glut4 to screen representative mutations using Taqman-PCR methods.
Results The 72.11% of subjects were confirmed to have OSAS.Detection of OSAS is significantly higher in those with FBG ≥ 5.6 mmol/L than in those with FBG<5.6 mmol/L(81.37% vs. 69.99%,
P=0.001).FBG ≥ 5.6 mmol/L is in significant independent relation with OSAS,even after adjusting for confounders(
OR=1.20,
CI:1.02-1.42,
P=0.026).rs5417 loci was successfully genotyped in 861 of the 870 subjects.Detection of OSAS was significantly higher in subjects with FBG ≥ 5.6 mmol/L than in those of FBG<5.6 mmol/L in carrier of AA+AC genotypes(81.11% vs.67.89%,
P=0.003),whereas no significant differences were observed in those with CC genotypes.Risk of OSAS was significantly higher in AA+AC carriers with FBG ≥ 5.6 mmol/L than in those with FBG<5.6 mmol/L,after controlling for age,gender,systolic and diastolic blood pressure,and lipids(
OR=1.88,95%
CI:1.16-3.05,
P=0.011).Risk for OSAS was significantly higher in carriers of CC genotype of those with FBG ≥ 5.6 mmol/L than in carriers of AA+AC genotypes with FBG<5.6 mmol/L(
OR=2.02,95%
CI:1.04-3.92,
P=0.038).
Conclusion CC genotypes of rs5417 locus in Glut4 gene and a FBG ≥ 5.6 mmol/L might interact to increase the risk of OSAS.
